Procedure: Chickens were immunized with GKPIPNPLLGLDST (V5 epitope) conjugated to KLH. After multiple immunizations in Freunds adjuvant, eggs were collected and the immunoglobulin fraction prepared. Antibodies were immunoaffinity purified using the peptide immobilized on a solid support and conjugated with horseradish peroxidase (HRP).
Specifications: By ELISA, the anti-V5 diluted 1:5,000 gave an O.D.= 1.0 in a 15 minute reaction against peptide conjugated to a different carrier than used for anti peptide purification. The Chicken anti-V5 was an effective probe at 1:1000 dilution.
