Procedure: Chickens were immunized with GKPIPNPLLGLDST (V5 epitope) conjugated to KLH. After multiple immunizations in Freunds adjuvant, eggs were collected and the immunoglobulin fraction prepared. Antibodies were immunoaffinity purified using the peptide immobilized on a solid support.
Specifications: ELISA: Antibody specificity was verified by ELISA against peptide conjugated to BSA (GKPIPNPLLGLDST-BSA). A 1:100,000 dilution of the antibody gave an OD=1.0 in a 15 minute reaction using HRP-conjugated Goat Anti Chicken IgY at 1:10,000 and TMB as the substrate. Appropriate specificity controls were run. Western Blots: Positope Marker ( Invitrogen Positope R900-50 ) was used for Western Blot evaluations at 100 and 25 ng. The anti V5 was an effective probe at 1:5000 dilution using HRP-affinity purified goat anti chicken IgY at 1:5000. BioRad Opti-4CN was used as substrate.
