Procedure: Chickens were immunized with MASMTGGQQMG (T7 epitope) conjugated to KLH. After multiple immunizations in Freunds adjuvant, eggs were collected and the immunoglobulin fraction prepared. Antibodies were immunoaffinity purified using the peptide immobilized on a solid support.
Specifications: ELISA: Antibody specificity was verified by ELISA against peptide conjugated to BSA (MASMTGGQQMG-BSA). A 1:60,000 dilution of the antibody gave an OD=1.0 in a 15 minute reaction using HRPO conjugated Goat Anti Chicken IgY at 1:10,000 and TMB as the substrate. Appropriate specificity controls were run. Western Blots: The anti T7 was an effective probe at 1:5,000 dilution using affinity purified goat anti chicken IgY HRPO at 1:5,000. BioRad Opti-4CN was used as substrate.
