Procedure:
Chickens were immunized with highly purified dog IgG isolated from normal dog serum. After multiple immunizations in Freunds adjuvant, eggs were collected and the IgY fraction prepared. The antibodies were immunoaffinity purified from a solid phase immunoabsorbent and conjugated to horseradish peroxidase (HRP).
Specifications:
In a direct ELISA using 1 ug/ml purified dog IgG as sample, conjugate diluted 1:25,000 produced an O.D.=1.0 in a 10 minute reaction with TMB as substrate. Optimal conjugate working dilutions should be determined for your particular assay conditions.
