Procedure:
Cytonectin was isolated from a cell lysate of normal human third trimester placenta by reversed-order OFarrell gel electrophoresis (ROOGE). Purified cytonectin was used to immunize a chicken. After multiple immunizations, eggs were collected and IgY was purified from the egg yolk.
Specifications:
For immunoblotting, antibody is diluted to a 1 mg/ml stock solution. This is diluted 1:1000 immediately prior to blot application. The bound primary antibody is detected by HRP-goat anti-chicken IgY Fc fragment as secondary antibody, followed by any chemiluminescence protocol. The anti-cytonectin antibody is also useful for immunomicroscopy. Most tissues have some cytonectin detectable. Many pathologic tissues, especially neoplastic cells, show increased cytonectin expression. The antigens are highly evolutionarily conserved in human, rodent, and avian tissues.
