Procedure:
Chickens were immunized with highly purified bovine serum albumin. After multiple immunizations in Freunds adjuvant, eggs were collected and the IgY fraction prepared. The antibodies were immunoaffinity purified from a solid phase immunoabsorbant and conjugated to horseradish peroxidase (HRP).
Specifications:
The anti BSA shows a single precipitin arc against bovine serum albumin and the albumin in bovine serum using immunoelectrophoresis. In a sandwich ELISA against purified bovine albumin with 20 ug/ml as sample, conjugate diluted 1: 3,000 produced an O.D.=1.2 in a 15 minute reaction with OPD as substrate. Optimal conjugate working dilutions should be determined for your particular assay conditions.
